What is Semax?
Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) analog of the ACTH(4-10) fragment, stabilized by a C-terminal Pro-Gly-Pro motif that confers resistance to enzymatic degradation. Originally developed in Russia, it is studied as a neurotrophic and nootropic research peptide and lacks the steroidogenic and melanocortin activity of full ACTH, isolating the neurotropic activity of the (4-10) fragment as a research tool.
Research areas where laboratories buy Semax for in vitro work include brain-derived neurotrophic factor (BDNF) and TrkB expression studies in hippocampal and basal-forebrain cultures, cerebral ischemia and stroke neuroprotection models in rodents, learning, memory and attention behavioral models, dopaminergic and serotonergic neurotransmission assays, and oxidative-stress and neuroinflammation models in cell culture. Semax is not approved for therapeutic use in the United States or European Union.

Semax Mechanism of Action
- Acts as a stable analog of the ACTH(4-10) fragment — the C-terminal Pro-Gly-Pro motif confers resistance to enzymatic degradation, extending activity relative to the native fragment.
- Upregulates brain-derived neurotrophic factor (BDNF) and its receptor TrkB in hippocampal tissue in rodent and cell-culture models.
- Modulates dopaminergic and serotonergic neurotransmission, the basis for much of the published neuropharmacology work.
- Inhibits enkephalin-degrading enzymes, prolonging the activity of endogenous regulatory peptides.
- Engages antioxidant and anti-inflammatory networks in models of cerebral ischemia and oxidative stress.
- Lacks the steroidogenic / melanocortin activity of full ACTH, isolating the neurotropic activity of the (4-10) fragment as a research tool.
Semax Research Applications
- BDNF and TrkB expression studies in hippocampal and basal-forebrain cultures.
- Cerebral ischemia and stroke rodent models examining neuroprotection endpoints.
- Learning, memory, and attention behavioral models in rodents.
- Dopaminergic and serotonergic neurotransmission assays.
- Oxidative-stress and neuroinflammation models in cell culture.
- Enzymatic-stability and structure-activity studies of the Pro-Gly-Pro–stabilized heptapeptide.
Why Buy Semax from Koi Peptides?
- Every batch of Koi Semax is synthesized and lyophilized in the United States, then released against independent third-party laboratory (Freedom Diagnostics Testing) testing.
- Each lot is tested by HPLC for purity, LC-MS for identity confirmation, and endotoxin assay for sterility readiness.
- The lot ID is printed on every vial and ties back to a public COA library, so researchers can verify documentation before they buy and after the vial arrives.
- Orders placed before 2 PM CT ship the same business day, and shipping is free on orders over $200.
